evaluation of cytogenetic alterations in peripheral blood lymphocytes of esophageal cancer patients treated with radiotherapy or chemoradiotherapy using cytokinesis-blocked micronucleus assay

چکیده
the effects of combined radiotherapy (rt) and chemotherapy in the severity of cytogenetic alterations expressed as micronucleus (mn) in peripheral blood lymphocytes of patients treated for esophageal cancer was evaluated. to do this, blood was obtained from 23 and 15 esophageal cancer patients scheduled for chemo-radiotherapy and rt alone, respectively, before, during, and after treatment. blood samples were cultured in rpmi-1640 complete medium containing 1% phytohemagglutinin and incubated in a co 2 incubator. cytochalasin-b was added to the cultures at a final concentration of 5μg/ml. finally, harvesting, slide making, and analysis were performed according to standard procedures. results indicate that there was no significant difference between the frequencies of mn in lymphocytes of individuals before being treated with rt alone or chemo-radiotherapy. in the middle of treatment, (after 12 fractions of rt) the frequency of mn increased significantly compared with their concurrent pre-treatment samples in both groups (four-fold). however, the frequency of mn observed for rt patients was not significantly different with those received chemo- and radiotherapy. at the end of treatment, (after 24 fractions of radiotherapy) an increase in the mn frequency was observed for chemo-radiation group significantly higher than rt group ( p =0.022). mild increase in mn frequency in lymphocytes of patients receiving chemoradiation only after the completion of treatment course might be indicative of resistance induced by chemotherapeutics to the clastogenic effects of radiation. therefore, using these agents repeatedly for cancer treatment in combination with radiation might not cause severe adverse biological effects in normal tissues.

Assessment of the radioprotective effects of amifostine on human lymphocytes irradiated in vitro by gamma-rays using cytokinesis-blocked micronucleus assay
Background: A radioprotective effect of amifostine as well as its ability to modulate the level of spontaneous and gamma-irradiation-induced genetic changes on human peripheral blood lymphocytes has been investigated . Amifostine, known as a potent radical scavenger, has been introduced as the most effective radioprotector, yet it is not completely approved for the clinical use. However, furt...

AIMS AND BACKGROUND Esophageal cancer consists of 5% of all cancers. In Eastern Anatolia region of Turkey, the rate was found to be 16%. The primary aim of the study was to evaluate effect of the localization and histologic type of tumor, stage of cancer and treatment modality, patient's sex, age and smoking status, Karnofsky performance score, and family history on survival. Background: Most of the cytogenetic dosimetry, except Premature Chromosome Condensation assay, saturates at 6 Gy. Present studies are aimed to assess feasibility of simple CB-MN assay for prescreening of high dose accidental exposures. Materials and Methods: Human peripheral blood lymphocytes from two donors were gamma irradiated to different doses ranging from 0-15Gy, cultured for 72 h. Cytoch...

The potential cytogenetic damage associated with pesticide use in Greek agricultural workers was evaluated using micronuclei (MN) as biomarkers in lymphocytes of peripheral blood and exfoliated cells of the buccal mucosa. In addition, the effects of pesticide exposure and other variables on the cytokinesis block proliferation index (CBPI) in lymphocytes were also evaluated. Both the exposed and...
متن کاملRadio-adaptive response of peripheral blood lymphocytes following bystander effects induced by preirradiated CHO-K1 cells using the micronucleus assay
Background: Radio-adaptive response and bystander effects are known phenomena occurring in cells following exposure to ionizing radiation (IR). In this study we examined possible radio-adaptation of lymphocytes following bystander effects induced by CHO-K1 cells. Materials and Methods: Whole blood and CHO-K1 cells were cultured in RPMI-1640 complete medium. Cells were separately irradiated with...
